It is our pleasure to mention Scientific Volume Imaging as a user example for our GATTA-Beads (soon available). SVI used ATTO 647N labeled nanobeads from GATTAquant for high resolution STED imaging. Images were acquired on a Leica TCS SP8 STED 3X system. By acquiring optimally sampled z-stacks, SVI was able to reconstruct 3D PSFs by averaging over multiple GATTA-Beads, followed by a PSF distilling process in the SVI Huygens Software. The distilled PSF was then used for deconvolution in the Huygens Professional Software. This is one of the first examples where a 3D STED PSF was accurately distilled from bead data and demonstrated to be beneficial to the STED deconvolution result.
Images were acquired with the Leica TCS SP8 STED 3X system at VUmc, Amsterdam, The Netherlands, and were processed with the Huygens Professional software.
